The Mühlendamm in Berlin is one of the main roads in the Mitte district and part of Bundesstraße 1. The name goes back to a dam created between 1220 and 1230, which, when expanded into a weir, operated several water mills. The bridge that has existed at this point since the 19th century to cross the Spree was named Mühlendammbrücke; it runs parallel to the Gertrauden Bridge.

The first forerunners of the Mühlendamm lock were built in the branch of the Spree in the Cöllnisches Stadtgraben in the 16th century.
The first navigable wooden lock was built at this point in 1578.
The Mühlendamm lock was built in 1894 as a single-chamber lock.
In 1942 the lock on Mühlendamm was converted into a two-chamber lock.
It overcomes a fall height of 1.51 meters.
