Elector Philipp Wilhelm (1685-90) provided the building site for the church building, stones from the thick tower of Heidelberg Castle, timber from the stately forests, sand from the city moat and free transport. The plan was drawn up by the architect Johann Adam Breunig (around 1660-1727). The first phase of construction lasted until 1723.
It was not until 1749 that construction work could be resumed thanks to the generosity of Elector Karl Theodor (1742-1799). The nave was roofed the following year, and the facade and vaulting were completed in 1751. However, the work dragged on until late autumn 1759. The Mannheim court architect Franz Wilhelm Rabaliatti (1716-1782) was in charge of construction.
After the Jesuit order was abolished in 1773, its tasks and institutions were taken over by the Lazarists from 1782 to 1793. In the following years the church served as a military hospital. Since November 1, 1809 it has been the parish church of the Catholic parish of the Holy Spirit.
The baroque furnishings are no longer preserved. The altar painting by Kaulbach student Andreas Müller has been preserved. Elector Frederick the Victorious is buried in a crypt in the northeast corner of the church.

The Jesuit Church is located in the immediate vicinity of the university in the heart of Heidelberg's old town. After the foundation stone was laid in 1711, the church building was completed in 1759. The church belonged to the adjoining Jesuit college: after the Jesuit order was abolished in 1773, the church initially served the lazarists and was later used for secular purposes (e.g. as a military hospital). In 1809 the Catholic parish of the Holy Spirit received the church as a parish church; until then, the Catholic and Protestant communities had shared the Protestant Church of the Holy Spirit. The interior of the Jesuit Church was last renovated in 2001-2004, with the altar island being redesigned. In 2009, the new large Kuhn organ was inaugurated, which in 2014 was joined by a small "sister", a baroque-style choir organ, also built by the Swiss organ building company Kuhn.
