The first documented mention of the Zeininger marca dates from the year 788 in the so-called Codex Laureshamensis of the Lorsch monastery. At that time, a man named Gubo donated a farm in Zainingen to the Lorsch monastery. Zainingen has belonged to Württemberg since 1383 and was assigned to the district of Urach. In 1938 the town came from the Oberamt Urach to the district of Münsingen. With the dissolution of the district of Münsingen as part of the district reform of 1973, the municipality came under the district of Reutlingen. On January 1, 1975, Zainingen merged with Böhringen and Donnstetten to form the new municipality of Römerstein.
