The Sophienkirche was built as a hall building. The foundation stone was laid on June 6, 1712, during the reign of King Frederick I of Prussia. After her husband's death on February 25, 1713, the originally intended namesake, Sophie Luise of Mecklenburg-Schwerin (1685–1735), Frederick's third wife, was highly unwelcome to the subsequent King Frederick William I of Prussia at the inauguration of the church. When it was inaugurated on June 18, 1713, the church was named Spandau Church after its location in the Spandau suburb. It was only under the successor King Friedrich II that it was given the originally intended name after Sophie Luise: Sophienkirche.
