The garrison church stands on the site of the Holy Spirit Church, which was mentioned for the first time in 1772. In 1376 and 1659, after being rebuilt in the Gothic style, the church burned down. The garrison church was then built for a Prussian garrison in 1738-1741.

A “House of the Holy Spirit” was mentioned in the same place as early as 1272. Around 1338, a hospital was built by monks from Greifswald; the associated church was originally intended for members of the hospital. After the Second Northern War, the previously burned down church was rebuilt as a garrison church to look after the Prussian soldiers. However, the Anklam and the Prussians were initially unable to agree on a name. The name “Garnissionkirche” was placed over the west entrance and “Heiligengeistkirche” over the south portal.
