This small chapel was built in 1457 by order of the Confraternity of Saints Anthony and James to allow pilgrims stopping at the nearby hospital to participate in religious ceremonies.

The oratory was originally the church of the pilgrim hospice, which housed pilgrims on their way to Rome. The magnificent paintings and frescoes by Matteo da Gualdo, Pierantonio da Foligno and Andrea d'Assisi, a pupil of Perugino, date from the 15th century. The chapel is dedicated to Saint Anthony and Saint James of Compostela, to whom the frescoes on the walls to the right and left of the altar are dedicated; behind the altar an interesting fresco by Matteo da Gualdo depicting the Madonna with Child and two holy pilgrims with their staffs.
