The Cathedral, dedicated to the Assumption, is a true religious, civic, and historical symbol of the city. It was Frederick II who commissioned its construction in 1232, in keeping with the Romanesque art and architecture of the time. Altamura Cathedral is one of the four Palatine basilicas of Puglia, meaning those directly dependent on the Emperor of Swabia. Much of the original church collapsed in the 1316 earthquake and was rebuilt during the reign of Robert of Anjou.

Altamura Cathedral, dedicated to S. Maria Assunta, was built by Frederick II, at the same time as the foundation of the city, between 1232 and 1254. Partially destroyed, perhaps by an earthquake, it was rebuilt in 1316.
A plaque walled up above the Angevin gate, protected by an iron gate, reminds us of the ruin of the Temple which took place under the government of Roberto D'Angiò. Its reconstruction was entrusted to the sons of a certain Consiglio di Bitonto, a master of art.
