The town church, which was built in the 14th century, was changed and redesigned many times. The Marienkirche with three-sided Ostschluss has a simple west gable, which was designed as a gable. The outer buttresses with the interspersed arched windows shape the appearance and supported the earlier Gothic vaults. Today the church has a pitched roof and inside a grooved flat ceiling.

The interior features the magnificent, wooden pulpit from 1587. It was built by the Lübeck-based Tönnies Evers d. J. originally made for the Marienkirche in Wismar and came in 1746 to Neustadt-Glewe. The pulpit had already been painted over in Wismar. The parapets with a Renaissance ornament are structured by imaginary columns. In the round-arched niches Christ and five apostles are depicted.

The wooden altar with its painting of the Deposition from the Cross, which today is attached separately to the south wall of the church, dates back to the Baroque period. The painter was Johann Heinrich Suhrlandt, it is a copy of an image by Anthonis van Dyck. The Gothic Neustädter Altar, originally from Lübeck, was made around 1420-1435 by the master of the Jakobialtar. He is since 1841 in the State Museum Schwerin.

The organ from 1873 was made by Friedrich Albert Mehmel and restored in 1996. The organ loft was built around 1770 in connection with the construction of the predecessor organ of Paul Schmidt (Rostock).

After the great city fire of 1728, the church had to be rebuilt on the foundation walls. The church bell is located in the half-timbered outbuilding built in the 18th century.

The Marienkirche has a gable roof and a flat ceiling inside. The bells are housed in an adjacent half-timbered building from the 18th century. The pulpit from 1587, which was only brought to Neustadt-Glewe from the Marienkirche in Wismar in 1746, is particularly worth seeing in the furnishings of the church. Christ and five apostles are also depicted in their arched niches.

The altar is on the south wall of the church. It is decorated with a painting of the Descent from the Cross. The old Gothic altar has been in the Schwerin State Museum since the middle of the 19th century. The organ dates from 1873 and was restored in 1996.
