Islamitische oorsprong: De oorspronkelijke poort, bekend als de Oude Bisagrapoort of Bab al-Saqra ("Heilige Poort"), werd gebouwd in de 9e eeuw tijdens de islamitische overheersing.
Renaissance-reconstructie: In de 16e eeuw, onder het bewind van Karel V en Filips II, werd de Nieuwe Bisagrapoort gebouwd, de poort die er vandaag de dag nog staat. De bouw begon rond 1540 en werd voltooid in 1576.
