The high school building was constructed between 1905 and 1907 according to plans by the architects Wellbrock and Schäfer on land formerly belonging to the Durlach Palace gardens. During construction, the remains of an ornamental fountain from the palace gardens were discovered. As early as 1911, the building was expanded with an additional structure designed by the architect Karl Kohler.

The high school was severely damaged in an air raid in 1944.

The schoolyard can be accessed from Karl-Weysser-Straße. There stands another building dating from around 1907, featuring a stylized tree on its gable end; this was the former caretaker's cottage.

The memorial plaque commemorating the two World Wars is located on the north wing in the schoolyard.

The Markgrafen-Gymnasium in Durlach shares its origins with the Bismarck-Gymnasium in Karlsruhe. Both educational institutions developed from the Gymnasium opened in Durlach in 1586 and shared a similar history until 1724. In 1724, Margrave Karl Wilhelm of Baden-Durlach relocated the majority of the Gymnasium to his newly founded residence in Karlsruhe, while the rest of the school continued as the "Pädagogium." Latin was again taught at the Pädagogium by 1781 at the latest. In 1837, it was merged with the Höhere Bürgerschule (Higher Citizens' School). With the move into the new building of the Vereinigte Schulen Durlach (United Schools Durlach), built according to plans by Heinrich Lang in 1877/78 (today the Friedrich-Realschule), the school was elevated to a Pro- and Realgymnasium (pro- and realgymnasium). From 1902, it was a Realgymnasium.
