The church was built from 1575 to accommodate the Jesuits who arrived in the city the previous year after Bernardino Realino da Carpi, who died in Lecce in 1616 and was subsequently canonized. The construction of the Church of Gesù involved the demolition of the ancient Church of San Niccolò dei Greci of the Greek Orthodox Rite. The construction of the building was carried out according to the designs of the Jesuit Giovanni De Rosis from Como. It was opened for worship as early as 1577, but work continued for several more decades.
