The church is dedicated to Pope Saint Sixtus II, who is considered a co-patron of Halberstadt Abbey. This suggests both a close connection to this monastery and an early origin of the church. According to a document from 1483, a first church building is mentioned in 1016. It is still unclear whether this stood at the current location and can be regarded as a predecessor building. Dendrochronological investigations of an anchor beam from the southern tower arm prove a shoring in the last third of the 11th century. (as of 2015). This makes St. Sixtus the oldest securely dated city church in Saxony-Anhalt.

The beautiful town church of St. Sixtus zu Ermsleben is located on the street of the village churches. It was first mentioned in the 1st century and is a complex from different building epochs that have not been clarified. Inside is a Renaissance epitaph from 1571 for the Counts of Hoym.
